PARTHENON THERAPEUTICS is discovering and developing novel anti-cancer medicines that are designed to reprogram the tumor microenvironment. Many cancers create immune exclusion barriers that prevent the immune system from attacking tumors. The Parthenon approach to overcoming these barriers is to modulate the interplay among cancer cells, stromal cells, and immune cells.
Our lead program, PRTH-101, is a first-in-class humanized monoclonal antibody that blocks the interaction of Discoidin Domain Receptor 1 (DDR1) with collagen, thereby "punching holes" in the extracellular matrix barrier created by the tumor. With the extracellular matrix barrier breached, immune cells are able to infiltrate and attack the tumor. PRTH-101 is poised to enter Phase 1 clinical trials.
PRTH-101 is followed by a pipeline of preclinical projects targeting orthogonal components of the tumor microenvironment that promote immune exclusion. Parthenon combines its drug discovery and development efforts with a precision oncology approach to get the right medicines to the right patients. We use proprietary AI technology to digitally deconstruct the TME and develop unique insights to enable biomarker-driven patient selection strategies designed to guide clinical applications.

Position Summary
We are seeking a self-motivated
Principal Scientist/Associate Director, Protein Science to lead the protein science efforts across Parthenon's drug discovery pipeline and help build the Protein Science group. The successful candidate will be responsible to develop and implement scientific strategies and workflows to generate high-quality proteins in support of research and development efforts.
Role & Responsibilities:
- Identify CROs to build out Parthenon's external protein science capabilities. Oversee and manage CRO activities to ensure production of high quality proteins and scalability of protein expression/purification/characterization efforts.
- Develop workflows for protein expression and purification, quality control, and functional validation in close collaboration with project teams
- Oversee in-house labeling of existing protein reagents and QC'ing and validation of tagged proteins
- Establish protein ordering and tracking system, and implement into protein science workflow, help build LIMS
- Develop plan and lead efforts to internalize protein science capabilities as appropriate
- Collaborate with Biotherapeutics colleagues to develop and implement analytical methods to support biophysical assessment of proteins and antibodies including binding affinity and kinetics assessments, structural validation, and developability assessments
- When appropriate, publish high-quality research relevant to the mission of Parthenon
Job Qualifications
- Degree in Biochemistry, Molecular/Cellular Biology, or a related field + indicated years of relevant industry experience in biologics drug discovery:
- Ph.D. + 3-7 yrs – or
- M.S. + 10-14 yrs – or
- B.S. + 12-16 yr
- Required:
- Expertise and hands-on experience with mammalian protein expression, including 293 and CHO-based systems and stable cell line generation; high-throughput expression screening and scale-up to multiliter shake flasks and WAVE bag systems
- Expertise and hands-on experience with state-of-the-art protein purification methods, including FPLC, HPLC; affinity-based, size exclusion, and ion exchange chromatography
- Experience managing external CROs
- Broad knowledge in protein characterization and validation methods, including SDS and native PAGE, Western blot, Surface Plasmon Resonance (SPR), BioLayer Interferometry (BLI), AlphaLISA, HTRF, ELISA, CD, and SEC MALS
- Strong interpersonal skills, integrity, and excellent verbal and written communication skills
- Demonstrated ability to excel in a fast-paced, cross-functional team environment
- Creative problem solver with demonstrated track record in production of difficult-to-express proteins and protein complexes
- Preferred:
- Structural biology experience, such as X-ray crystallography or HDX
- High-throughput antibody expression and purification experience
- Experience with antibody developability assessment, including HIC, AC SINS, polyspecificity (BVP ELISA and others), stability determination
